Paula Tamagnini | Cyanobacterial EPS: From the genes to the industrial toolbox

Paula Tamagnini - University of Porto, Portugal Homepage: https://www.i3s.up.pt/research-group.... Cyanobacterial extracellular polymeric substances (EPS): From the genes to the industrial toolbox Many cyanobacteria produce extracellular polymeric substances (EPS) mostly composed of heteropolysaccharides. These EPS have distinctive characteristics that make them suitable for biotechnological/biomedical applications. However, the information on their biosynthetic pathways is still limited hindering their manipulation/optimization. A phylum-wide analysis led to the identification of genes/proteins putatively involved in the assembly and export of EPS and revealed that cyanobacterial EPS production may not strictly follow one of the pathways previously characterized for other bacteria. The generation and characterization of several mutants by us and others allowed to pinpoint genes/proteins involved in EPS production and to determine the function of key proteins homologs to bacterial components of the Wzy- and ABC transporter-dependent pathways. In parallel, the methodology for the production and isolation of these cyanobacterial polymers was optimized, the polymers were characterized and tested for several applications. The unusual features of the polymers endorse its use for the removal of metal cations from aqueous solutions, vehicles for controlled delivery of functional proteins and vitamins, the development of coatings with anti-adhesive properties, wound-healing, and as antitumor agents.
