Masson's Trichrome Staining method is used for differential staining of collagen fibers and smooth muscular fibers in tissues such as skin, heart, etc. on formalin-fixed, paraffin-embedded sections, and may be used for frozen sections as well. As a result nuclei will be stained in black color, cytoplasm, keratin, muscles – red, collagen – blue, erythrocytes – yellow. Masson's Trichrome Staining Protocol: 1. Weigerts hematoxylin – 10 min. 2. Dry slides without washing. 3. Alcoholic solution of picric acid – 4 min. 4. Wash the slides quickly in distilled water (3 - 4 sec). 5. Acid crimson fuchsin solution – 4 minutes. 6. Wash the slides in distilled water. 7. Phosphomolybdic acid – 10 min. 8. Dry slides without washing. 9. Massons anyline blue – 5 min. 10. Wash slides in distilled water. 11.
