The primer in cellular DNA replication is an RNA molecule was proved when an M13 phage DNA was allowed to replicate in E. coli cell lysate in presence of the antibiotic rifampicin. Replication was inhibited in presence of rifampicin which is an inhibitor of RNA polymerase. This gave the first indication that RNA polymerase (primase) is involved in the process of DNA replication. The RNA polymerase primes the process with a short RNA synthesis (primer). DNA polymerase can not start denovo because it has got a proofreading activity which RNA polymerase lacks. A proofreading polymerase cannot start denovo synthesis. RNA polymerase does not require proofreading because error in RNA synthesis is most of the time tolerated, as most of the missence and silent mutations do not seriously affect the function of the proteins. Even if it does it will only affect that protein which is synthesized from that particular incorrect RNA. DNA being genetic material its accurate synthesis is essential.The primer is definitely an RNA molecule was further proved when Okazaki fragments could not be completely digested with DNase.
