There are two types - insertional and replacement - of vectors derived from lambda bacteriophage. This requires the use of spi phenotype since lambda phage vectors have no selection or screening markers. From the lambda bacteriophage vector, cosmids are created, relying on the unique function of cos sites. Understanding cosmid means understanding the structure and function of cos sites - a short section of this video explains the anatomy and function of cos sites in lambda phage and how it is used in lambda phage vector. Cosmid containing lambda phage viruses are assembled in vitro by extracting lambda phage proteins from bacteria and mixing it with the cosmid DNA in a tube. Lambda phage vectors and cosmids have specific complications when it comes to their application - most significant is the problem of cosmid concatermization and we discuss it towards the end of this video on lambda phage vectors.
